A short duration pulse of polarized light is used to excite the sample. The decay of fluorescence is monitored first paralell (I||(t)), then perpendicular (I-(t)) to the excitation. From these measurements one can determine the decay of anisotropy r(t)
Anisotropy measurements allow one to determine how far a species rotates during its fluorescecne lifetime.
